Prothrombin Time in Dicoumarol Therapy.
نویسنده
چکیده
In the course of controlling dicoumarol therapy given (a) as a postoperative routine to prevent thrombosis (fifty-five cases) and (b) therapeutically in cases of established venous thrombosis (nine cases), certain peculiarities have been observed in the behaviour of the prothrombin time, particularly in relation to dicoumarol haemorrhage. Two sources of thromboplastin have been used, Russell viper venom and human brain. Data are presented which suggest that the brain extract is to be preferred to venom, for estimations of prothrombin levels are then more in accord with clinical observation. Technical Methods

The action of p-aminosalicylic acid on prothrombin time in man.
A lengthening in prothrombin time by giving salicylates was first observed in the animal experiments of Link, Overman, Sullivan, Huebner, and Scheel (1943). This result, and a comparison of the structures of coumarin-like anticoagulants (Overman, Stahmann, Huebner, Sullivan, Spero, Doherty, Ikawa, Graf, Roseman, and Link, 1944), led Link to conclude that dicoumarol (3,3'-methylenebis [4hydroxyc...
متن کاملVitamin K and the biosynthesis of prothrombin. II. Structural comparison of normal and dicoumarol-induced bovine prothrombin.
Highly purified dicoumarol-induced bovine prothrombin, which does not bind calcium ions and has no prothrombin activity, has been structurally compared with normal prothrombin. Quantitative amino acid and carbohydrate analysis gave identical results for both prothrombins, as did analysis of the NHz-terminal and the COOH-terminal amino acids and molecular weight determination with the sodium dod...
متن کاملVitamin K and the biosynthesis of prothrombin. IV. Isolation of peptides containing prosthetic groups from normal prothrombin and the corresponding peptides from dicoumarol-induced prothrombin.
The 25,000 Af, fragment obtained by thrombin digestion of normal prothrombin has been further compared with the corresponding fragment from dicoumarol-induced prothrombin. In contrast with the fragment from normal prothrombin, the one from dicoumarol-induced prothrombin does not bind calcium (STENFLO, J. (1973) /. Biol. Chem. 248, 6325). After cyanogen bromide degradation of this material, a fr...
متن کاملThe progress of prothrombin time measurement
Warfarin is the most widely used medicine for oral anticoagulant therapy (OAT). It inhibits the synthesis of coagulation factors II, VII, IX, and X in the liver and results in the production of inactive or partially active versions of these factors. Inactive coagulation factors interfere with prothrombin time measurement (Quick and Owren PT) measuring the sum of coagulation activity and inhibit...
